La formaci\u00f3n de este \u00f3rgano incluye una serie de etapas: infecci\u00f3n de los pelos radicales por la bacteria, formaci\u00f3n del cord\u00f3n de infecci\u00f3n, formaci\u00f3n de un meristemo nodular, y diferenciaci\u00f3n de la bacteria en bacteroides, la forma fijadora de nitr\u00f3geno. La interacci\u00f3n entre la bacteria y la planta origina una regulaci\u00f3n espacial y temporal de una serie de genes vegetales, las nodulinas. Durante las etapas tempranas de la infecci\u00f3n, las paredes de las c\u00e9lulas vegetales deben ser hidrolizadas de una manera espec\u00edfica y controlada, para permitir la penetraci\u00f3n de la bacteria. La degradaci\u00f3n de las paredes vegetales libera una serie de compuestos reguladores de otros genes de la planta. Este trabajo se ha centrado en determinar la importancia de las enzimas p\u00e9cticas de medicago en los primeros estadios de la interacci\u00f3n simbi\u00f3tica con sinorhizobium meliloti. En primer lugar, se ha demostrado la expresi\u00f3n espec\u00edfica de una poliglacturonasa, mspg3, en la zona de infecci\u00f3n del n\u00f3dulo. Asimismo, se ha identificado y secuenciado la regi\u00f3n promotora de este gen, y se ha comprobado su funcionalidad en el sistema heter\u00f3logo vicia hirsuta-rhizobium leguminosarum. La regi\u00f3n 3\u00c2\u00bfno codificante de mspg3 presente una secuencia que se encuentra  muy repetida en el genoma de medicago. Por otra parte, se han identificado otros genes p\u00e9cticos cuya expresi\u00f3n est\u00e1 asociada m\u00e1s o menos espec\u00edficamente al desarrollo del n\u00f3dulo. Las especies de medicago estudiadas poseen dos genes de pectato liasas distintos, pl1 y pl2, cuya expresi\u00f3n de detecta asociada a la simbiosis y al desarrollo de la flor. Asimismo, existen en medicago diversos genes de pect\u00edn metilesterasas, cinco de los cuales se expresan durante la nodulaci\u00f3n.
