El gran tama\u00f1o del genoma del vgpt ha dificultado hasta muy recientemente la obtenci\u00f3n de un cdna infectivo de longitud completa que facilite la manipulaci\u00f3n g\u00e9nica del virus. Por ello, en el laboratorio nos hemos centrado en el aislamiento y caracterizaci\u00f3n de rnas defectivos, minigenomas, de menor longitud que el genoma parental. En esta tesis se han puesto a punto dos sistemas de expresi\u00f3n de minigenomas sint\u00e9ticos rna derivados del vgpt bajo los promotores t7 y de citomegalovirus (cmv). estos minigenomas son replicados y empaquetados (es decir, rescatados) en presencia de virus complementador. Mediante la obtenci\u00f3n de mutantes de deleci\u00f3n de los minigenomas y el analisis de la expresi\u00f3n del gen trazador de la b-glucuronidasa, se han determinado las secuencias minimas necesarias para su replicaci\u00f3n. Estas secencias se han reducido a 1164 y 278 nucleotidos en los extremos 5\u00c2\u00bf y 3\u00c2\u00bf del rna viral, respectivamente.  el minigenoma m\u00e1s peque\u00f1o que ha sido rescatado en presencia de virus complementador tiene 3.3 kb (m33), mientras que los minigenomas m\u00e1s peque\u00f1os que son replicados pero no encapsidados tienen 2.1-2.2 kb (m21 y m22). La diferencia entre los minigenomas en viriones del vgpt est\u00e1 en estudio.
