{"id":87181,"date":"2000-03-11T00:00:00","date_gmt":"2000-03-11T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/bioquimica-de-las-reacciones-de-tranferencia-de-cadenas-de-dna-catalizadas-por-la-proteina-trwc\/"},"modified":"2000-03-11T00:00:00","modified_gmt":"2000-03-11T00:00:00","slug":"bioquimica-de-las-reacciones-de-tranferencia-de-cadenas-de-dna-catalizadas-por-la-proteina-trwc","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/bioquimica-de-las-reacciones-de-tranferencia-de-cadenas-de-dna-catalizadas-por-la-proteina-trwc\/","title":{"rendered":"Bioquimica de las reacciones de tranferencia de cadenas de dna catalizadas por la proteina trwc"},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Hernando Mart\u00edn Miguel \u00e1ngel <\/strong><\/h2>\n<p>En este trabajo se analizan a nivel bioqu\u00edmico las reacciones de transferencia de cadenas de dna que cataliza la prote\u00edna trwc y m\u00e1s concretmaente, un dominio de la prote\u00edna bien definido, donde reside la actividad relaxasa.  trwc es el enzima que se une al origen de transferencia y cataliza el corte sitioespec\u00edfico del dna que inicia y termina la transferencia del dna en el proceso conjugativo.  se ha comprobado que la regi\u00f3n aminoterminal de trwc puede considerarse como un dominio, estructural y funcionalmente independiente del resto de la prote\u00edna, localizado entre el extremo aminoterminal de la prote\u00edna y el residuo de arginina r300.  el dominio aminterminal es capaz de llevar a cabo las reacciones que in vitro, caracterizan la actividad relaxa de un enzima: uni\u00f3n de ssdna, corte y transferencia de ssdna y relajaci\u00f3n de dsdna. Estas reacciones son espec\u00edficas de secuencia, siendo el \u00fanico sustrato v\u00e1lido el sitio nic de r388 y no el de otros sistemas conjugativos.  la estructura nativa del dominio aminoterminal de trwc se recupera espont\u00e1neamente, en las condiciones adecuadas, tras denaturalizar la prote\u00edna con un tratamiento t\u00e9rmico. Al igual que la estructura, la actividad relaxa, medida a trav\u00e9s de las reacciones mencionadas en elpunto anterior, tambi\u00e9n se recupera.  el segmento comprendido entre los residuos 275-293
