{"id":87256,"date":"2000-09-11T00:00:00","date_gmt":"2000-09-11T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/identificacion-i-caracterizacion-de-acuaporinas-en-vitis-richter-110\/"},"modified":"2000-09-11T00:00:00","modified_gmt":"2000-09-11T00:00:00","slug":"identificacion-i-caracterizacion-de-acuaporinas-en-vitis-richter-110","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/identificacion-i-caracterizacion-de-acuaporinas-en-vitis-richter-110\/","title":{"rendered":"Identificacion i caracterizacion de acuaporinas en vitis (richter-110)"},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Baiges Blanco Isabel M. <\/strong><\/h2>\n<p>Las acuaporinas o canales de agua son unas proteinas de expresion ubicua cuya funci\u00f3n es la de facilitar el transporte de agua a favor de gradiente a trav\u00e9s de las membranas biol\u00f3gicas.  en esta tesis doctoral se ha estudiado la expresi\u00f3n de acuaporinas en el h\u00edbrido richter-110(vitis berlandieri x vitis rupestris) por su mayor resistencia a la sequia posiblemente relacionada con el vigoroso crecimiento radicular.  mediante rt-pcr se obtuvieron sondas especificas para la selecci\u00f3n de clones positivos en una biblioteca de cdna obtenida a partir de mrna de hoja de vitis richter-110. El resultado fue la deteccion de 7 clones distintos de secuencia codificante completa y uno con la secuencia codificante parcial. las secuencias nucleot\u00eddicas completa y uno con la secuencia codificante parcial. Las secuencias nucleot\u00eddicas y polipept\u00eddicas pueden obtenerse de la base de datos genbank. El analisis de la actividad transportadora se llev\u00f3 a cabo mediante la expresi\u00f3n heter\u00f3loga en oocitos de xenopus laevis y demostr\u00f3 que al menos tres de las proteinas analizadas son acuaporinas.  el estudio de la inducibilidad a nivel transcripcional desvel\u00f3 que dos de estos genes se regulan siguiendo ciclos circadianos siendo la expresi\u00f3n mayor durante la primera parte del d\u00eda. La expresi\u00f3n genica de estos mismos dos genes en respuesta al estr\u00e9s h\u00eddrico fue ligeramente distinta a los valores control. Mediante la utilizaci\u00f3n de anticuerpos policlonales se determin\u00f3 el efecto del estr\u00e9s hidrico y del acido abscisico(aba) en la expresi\u00f3n de esta proteinas. El analisis de la expresi\u00f3n genica a nivel transcripcional en distintos tejidos de la planta se realiz\u00f3 mediante northern inverso (dot-blot). Dos de los genes analizados se expresan predominantemente en tejidos radiculares.
