{"id":87414,"date":"2018-03-10T00:12:12","date_gmt":"2018-03-10T00:12:12","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/aplicaciones-citometricas-al-ciclo-bacteriano\/"},"modified":"2018-03-10T00:12:12","modified_gmt":"2018-03-10T00:12:12","slug":"aplicaciones-citometricas-al-ciclo-bacteriano","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-de-la-vida\/aplicaciones-citometricas-al-ciclo-bacteriano\/","title":{"rendered":"Aplicaciones citom\u00e9tricas al ciclo bacteriano"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Jaume F Comas Riu <\/strong><\/h2>\n<p>Para valorar y aplicar los sistemas de citometria de flujo convencionales al an\u00e1lisis bacteriano se han aplicado t\u00e9cnicas de an\u00e1lisis citom\u00e9trico a un conjunto de situaciones y tratamientos en cultivos ax\u00e9nicos bacterianos. las medidas se han hecho sobre la part\u00edcula respecto a criterios de tama\u00f1o y recuento celular, analizando las se\u00f1ales obtenidas con diferentes marcadores fluorescentes.  la aplicabilidad de la t\u00e9cnica ha sido demostrada en la diferenciaci\u00f3n de las diferentes fases del ciclo bacteriano analizando par\u00e1metros citom\u00e9tricos estructurales y funcionales. A continuaci\u00f3n se realiza la comparaci\u00f3n citom\u00e9trica entre diferentes cepas en estado estacionario, y el an\u00e1lisis estad\u00edstico de sus distribuciones con la finalidad de encontrar unos rasgos caracter\u00edsticos para cada cepa o grupo.  la cuantificaci\u00f3n y an\u00e1lisis del mecanismo de acci\u00f3n de diferentes agentes letales y t\u00f3xicos son estudiados citom\u00e9tricamente en dos organismos (e.Coli y s.Aureus), de caracter\u00edsticas biol\u00f3gicas y citom\u00e9tricas bien diferenciadas. se aplican en estas cepas tratamientos de referencia de efectos conocidos (choque t\u00e9rmico, paraformaldehido y gramicidima) y se analiza por citometria su efecto seg\u00fan las variaciones detectadas en los par\u00e1metros de refrigencia celular y usando marcadores del contenido aparente de \u00e1cidos nucleicos, permeabildiad de membrana y actividad metab\u00f3lica (fundamentalmente potencial de membrana y actividad ester\u00e1sica intracelular). Una vez establecidos los efectos citom\u00e9tricos causados por estos tratamientos, se aborda el an\u00e1lisis de tratamientos m\u00e1s complejos que act\u00faan sobre la c\u00e9lula a diferentes niveles, como la aplicaci\u00f3n de tensioactivos y el choque osm\u00f3tico. El an\u00e1lisis citom\u00e9trico de estas sitauciones permite detectar las c\u00e9lulas a pesar de la presencia de part\u00edculas abi\u00f3ticas en el rango de tama\u00f1o celular, y hacer medidas
