Pilar <\/strong><\/h2>\n<p>Esta tesis describe la clonaci\u00f3n molecular de varias rips(\u00abribosome-inactivating proteins\u00bb) y lectinas presentes en sambucus nigra l. Y muscari armeniacum l. Miller, utilizando la t\u00e9cnica denominada race (\u00abrapid amplification of cdna ends\u00bb). El sa\u00faco (s. Nigra) posee un gran n\u00famero de rips en diversos tejidos, tanto de tipo 1 como de tipo 2, as\u00ed como numerosas lectinas. Con este trabajo se ha conseguido la clonaci\u00f3n de la nigrina 1,rip de tipo 2 no t\u00f3xica presente en las hojas de la planta, asi como de dos lectinas del mismo tejido, una monom\u00e9rica(snalm) y otra dim\u00e9rica(snald). Esta \u00faltima es la primera lectina dim\u00e9rica aislada en el genero sambucus. El gen de la nigrina 1 incluye la secuencia de un p\u00e9ptido se\u00f1al y de un p\u00e9ptido de conexi\u00f3n entre sus dos cadenas constituyentes, que son procesados en la maduraci\u00f3n de la prote\u00edna. Posee una alta homolog\u00eda con el gen de la rip de tipo 2 de corteza nigrina b. Los genes de las lectinas incluyen secuencias equivalente al p\u00e9ptidos se\u00f1al, parte de la cadena a y p\u00e9ptido de conexi\u00f3n de una rip de tipo 2, que se procesan para rendir las prote\u00ednas maduras. este hecho, junto con la elevada homolog\u00eda que presentan con la cadena b de la nigrina 1, hace pensar en un posible origen com\u00fan de todos estos genes.  as\u00ed mismo, se han obtenido los cdnas correspondientes a cuatro rips de tipo 1 presentes en los bulbos de la especie muscari armeniacum l. Miller, llamadas musarminas, asi como a otra musarmina presente en las hojas. Por comparaci\u00f3n con el tama\u00f1o de las prote\u00ednas nativas se deduce la posibilidad del procesamiento de un p\u00e9ptido(p\u00e9ptido se\u00f1al) en n-terminal asi como en el extremo c-terminal.  posteriormente se llev\u00f3 a cabo la expresi\u00f3n en escherichia coli de uno de los genes de musarminas de bulbo y la puesta a punto de la purificaci\u00f3n de la prote\u00edna recombinante.
