Adem\u00e1s, la rapida recuperaci\u00f3n de granulocitos y plaquetas disminuye la morbimortalidad relacionada con el procedimiento. la hemopoyesis puede mantenerse in vitro aplicando unas condiciones de cultivo especificas. Mediante la utilizacion de citoquinas selectivas se pueden generar celulas in vitro con caracteristicas funcionales definidas, en  unos cultivos llamados de expansi\u00f3n.  hipotesis: la coinfusi\u00f3n de ph obtenidos mediante expansi\u00f3n reducir\u00e1 el tiempo de neturopenia tras trasplante de scu.  objetivos: la evoluc\u00f3n de la eficacia, aplicabilidad y resultados de un metodo de generacion de ph mediante expansion.  resultados: la amplificaci\u00f3n de ph es significativamente mayor partiendo de c\u00e9lulas seleccionadas que de celulas monocucleadas. La adici\u00f3n de citoquinas de acci\u00f3n temprana, de acci\u00f3n intermedia y\/o tardia promueve  una mayor generaci\u00f3n de ph comprometidos. En cultivos de 6 dias, existe un mayor rendimiento si se adicionan citoquinas diariamente, y\/o se utilizan formas glicosiladas que aumentan su vida media. Es posible substituir la funci\u00f3n del suero de ternera fetal utilizado medios libres de suero comerciales y se disminuye el coeficiente de variaci\u00f3n (107% frente 61%). Mediante el an\u00e1lisis de las cafc, se ha podido de terminar que la combinaci\u00f3n m\u00e1s beneficiosa incluye scf, flt3-l, il6,il3 y mip-1a(recuperaci\u00f3n del 95%de cafc a los 6 dias de cultivo). La substituci\u00f3n de il3 y mip por tpo mejora los resultados y permite recuperar hasta un 145% de cafc. Se ha adaptado un m\u00e9todo de uso clinico (isolex-300i de baxter) para peque\u00f1as muestras de scu criopreservadas. Tras 6 dias de cultivo en medio libre de suero en presencia de scf,flt3-l,il6 y tpo, las celulas cd34+ iniciales se incrementan 7 veces.
