En la fase de iniciaci\u00f3n de la traducci\u00f3n intervienen una serie de factores proteicos denominados factores de iniciaci\u00f3n o eifs. Uno de ellos es el eif2 cuya fosforilaci\u00f3n en el residuo ser51 de su subunidad a tiene como consecuencia la inhibici\u00f3n de la sintesis general de proteinas. Existen cuatro tipos de eif2a quinasas que se activan en diferentes condiciones de estr\u00e9s celular. Una de estas eif2a quinasas es el hri (inhibidor regulado por hemina) que ha sido caracterizado en reticulocitos de conejo y se activa a bajas concentraciones de hemina. en esta tesis se ha purificado una actividad eif2a quinasa inhibible por hemina a partir de extractos de h\u00edgado de rat\u00f3n. Tambi\u00e9n se clon\u00f3 el hri de raton (mhri) y se demostr\u00f3 mediante anticuerpos espec\u00edficos que la actividad eif2a quinasa antes descrita corresponde al mhri. Mediante coexpresi\u00f3n de los mhris silvestre y mutante k196r en c\u00e9lulas de mam\u00edfero 293, y fusionados a dos ep\u00edtopos distintos (ha y flag), se demostr\u00f3 que el mhri forma in vivo homod\u00edmeros, que el mhri se expresa en una gran variedad de tejidos y en c\u00e9lulas nih-3t3.  tambi\u00e9n se purific\u00f3 y caracteriz\u00f3 una actividad inhibidora de eif2a quinasas (mki) a partir de los extractos de h\u00edgado de rat\u00f3n.  se ha clonado y caracterizado las dos primeras eif2a quinasas procedentes de la levadura schizosaccheromyces pombe, sek1 y sek2, que guardan gran similitud con el hri de mamiferos. Las proteinas recombinantes fosforilan especificamente la ser51 del eif2a. La actividad eif2a quinasa de sek1 y sek2 se inhibe por hemina a pesar de car\u00e1cter de los motivos de regulaci\u00f3n por hemina (hrm1 y hrm2) descritos en el hri.Sek1 parece estar regulada en s. Pombe por proteinas de choche t\u00e9rmico.
