La mol\u00e9culaha sido secuenciada completamente, presentando un tama\u00f1o de 15390 pb. Pt3.2i posee una estructura dim\u00e9rica compuesta por dos unidades pr\u00e1cticamente id\u00e9nticas e 6.4 kpb denominadas g\u00e9minis1 y g\u00e9minis2. Adicionalmente, contiene una secuencia de inserci\u00f3n denominada ist3. En funci\u00f3n del an\u00e1lisis de homolog\u00eda de secuencia, las unidades g\u00e9minis presentan una regi\u00f3n de replicaci\u00f3n muy similar a la de los pl\u00e1smidos de la familia cole2 y el gen deuna posible enzima alcohol deshidrogenasa(gen adh).  la regi\u00f3n de replicaci\u00f3n de pt3.21 posee el gen rnal, que probablemente codifica un rna antisentido, los genes repa y repb, que pueden codificar sendas prote\u00ednas de replicaci\u00f3n y el origen vegetativo de replicaci\u00f3n, ori. La prote\u00edna repa es capaz de unirse especificamente a la regi\u00f3n del dna que contiene la secuencia ori y es, probablemente, la prote\u00edna iniciadora de la replicaci\u00f3n.  la secuencia de inserci\u00f3n ist3, hom\u00f3loga a una serie de elementos m\u00f3viles descritos en diferentes bacterias, presenta copias en el cromosoma de thiobacillus t3.2 y en otros pl\u00e1smidos de la bacteria. Esta secuencia es capaz de mediar su transposici\u00f3n en c\u00e9lulas de e. Coli, donde se comprob\u00f3 que era responsable de varios reordenamientos plasm\u00eddicos.  el gen adh, que se expresa activamente en las c\u00e9lulas de thiobacillus t3.2, codifica un enzima perteneciente al grupo i de alcohol deshidrogenasas de cadena larga dependientes de zinc y nad(p).
