La tetx al igual que la bont\/a producen esta inhibici\u00f3n despu\u00e9s de una preincubaci\u00f3n a 37\u00c2\u00ba c a tiempos largos (horas). Esta inhibici\u00f3n de la liberaci\u00f3n de serotonina est\u00e1 bloqueda por los inhibidores de la actividad metaloprote\u00e1sica. Adem\u00e1s, la preincubaci\u00f3n a corto tiempo produce diferentes comportamientos por estas neurotoxinas indicadas:tetx produce una inhibici\u00f3n non competitiva de la captaci\u00f3n de serotonina en mayor\u00eda de las regiones de snc de rata, mientras que la bont\/a produce un ligero aumento de la captaci\u00f3n. Hemos encontrado que la tetx produce una inhibici\u00f3n de dosis- y de tiempo-dependiente de la captaci\u00f3n dependiente de na+ de [3h]serotonin,5-ht. Los inhibidores del transporte de serotonina tales como son fenfluramina (ic50,11.0+-0.9um), paroxetina(ic50,33.5+-0.1 um) e imipramina(ic50,89.9+-5.7 um), son de 3 o 4 ordenes menos potente que la tetx(ic50,8.7+-1.0 nm) encontrado una inhibici\u00f3n independiente de la temperatura en la preincubaci\u00f3n. La uni\u00f3n de [3h] imipramine y la ic50 de la fenfluramina no se afectan depues del tratamiento con la tetx, lo que indica que la tetx no act\u00faa directamente en el sitio de alta afinidad de serotonina al transportador de serotonina (sert) ni afecta el sitio donde act\u00faa la fenfluramina. Los fragmentos con actividad metaloprote\u00e1sica, l-tetx y l-bont\/a, son incapaces de producir el bloqueo de la captaci\u00f3n de serotonin, mientras que el fragmento hc-tetx y l-bont\/a, son incapacidades de producir el bloqueo de la captaci\u00f3n de serotonina con dosis dependiente (la ic50 varia entre 0.62 a 2.08 nm). Los inhibidores de la actividad metaloprote\u00e1sica que fueron capaces de bloquear la inhibici\u00f3n de la liberaci\u00f3n tanto basal como estimulada por k+ de [3h]5-ht en sinaptosomas de rata,
