Elena Miranda Ba\u00f1os <\/strong><\/h2>\n<p>El \u00f3rgano subcomisural(osc) es una gl\u00e1ndula del cerebro de todos los vertebrados, localizada en el techo del tercer ventr\u00edculo, cuyas c\u00e9lulas secretan glucoprote\u00ednas de alto peso molecular al l\u00edquido cefalorraqu\u00eddeo, donde polimerizan y forman la fibra de reissner(fr),que atraviesa el acueducto cerebral, el cuarto ventr\u00edculo y el canal central de la m\u00e9dula espinal. Hasta este trabajo de tesis doctoral, se dispon\u00eda de anticuerpos monoclonales anti-fr, con los cuales se desarroll\u00f3 un elisa sandwich para la detecci\u00f3n y cuantificaci\u00f3n de la secreci\u00f3n del osc.  como primer objetivo de esta tesis doctoral, se ha puesto a punto un protocolo de purificaci\u00f3n por cromatograf\u00eda de inmunoafinidad con dos de los anticuperos monoclonales anti-fr, con el que se ha logrado purificar casi a homogeneidad la secreci\u00f3n intracelular del osc de vaca. Las tres bandas purificadas, indentificadas en electroforesis y anteriormente descritas en la bibliograf\u00eda como de 540,450 y 320 kda, sehan utilizado conjuntamente como ant\u00edgeno para la generaci\u00f3n de un nuevo grupo de anticuerpos monoclonales anti-osc, as\u00ed,se han establecido quince l\u00edneas de hibridomas productoras de quince anticuerpos monoclonales, que se han caracterizado por inmunocitoqu\u00edmica, varios tipos de elisa e immunoblot. En inmunocitoqu\u00edmica, todos los anticuerpos monoclonales reconocen el osc y la fr de vaca, y la mayor\u00eda de ellos reaccionan con el osc de conejo y cerdo; por primera vez, se han obtenido adem\u00e1s dos anticuerpos que reconocen con intensidad el osc de rata y de rat\u00f3n. La caracterizaci\u00f3n por elisa ha mostrado que los anticuerpos monoclonales generados reconocen con alta afinidad las glucoprote\u00ednas del osc y la fr(elisa mediado por ant\u00edgeno), reaccionando con epitopos diferentes entre s\u00ed(elisa competitivo), que no se repiten y que coexisten(elisa sandwich) en las glucoprote\u00ednas de secreci\u00f3n.
