Belen <\/strong><\/h2>\n<p>Se han originado una serie de mutaciones espec\u00edficas en la secuencia de la fosfotructokinasa no alost\u00e9rica de d.Discoideum con objeto de investigar los motivos estructurales por los que carece de propiedades reguladoras. la mayor parte de los amino\u00e1cidos propuestos como importantes para el centro catal\u00edtico y sitios alost\u00e9ricos est\u00e1n conservados en esta pkk, excepto algunos de ellos cuya reversi\u00f3n no alter\u00f3 la conducta cin\u00e9tica del enzima nativo. Sin embargo, la introducci\u00f3n de diversas modificaciones a nivel de la cola c-terminal, \u00fanica para esta pfk, convirti\u00f3 a este isozima en uno alost\u00e9rico que exhibe propiedades caracter\u00edsticas de las pfks reguladoras, como hist\u00e9resis, intensa cooperatividad positiva para el fructosa 6-p, fuerte activaci\u00f3n por fructosa-2,6-p2 y fructosa-1,6-p2, inhibici\u00f3n por protones, dependencia de la concentraci\u00f3n de enzima y asociaci\u00f3n-disociaci\u00f3n de subunidades. La regi\u00f3n implicada ha sido caracterizada mediante mutag\u00e9nesis sistem\u00e1tica y el estudio de los mutantes ha suminsitrado una nueca interpretaci\u00f3n para el control alost\u00e9rico de las pkfs en general. Adicionalmente, hemos identificado a la pfk de d.Discoideum como un potente factor que inhibe la velocidad de polimerizaci\u00f3n de tubulina con una raz\u00f3n molar de una mol\u00e9cula por alrededor de 300 d\u00edmeros de tubulina para un efecto mitadl del m\u00e1ximo (ic50 = 32 nm). Este efecto ha sido caracterizado y es espec\u00edfico, puesto que no lo produce la pkf de m\u00fasculo. Tambi\u00e9n se contra de \u00e9sta \u00faltima, la actividad catal\u00edtica del enzima de d.Discoideum no se afecta por la presencia de tubulina o microt\u00fabulos.
