Mar <\/strong><\/h2>\n<p>El virus vaccinia tiene dos formas infecciosas distintas, una forma intracelular que s\u00f3lo sale al exterior por rotura mec\u00e1nica de la c\u00e9lula (imv), y una forma extracelular (eev), con una envuelta adicional derivada del tgn. El transporte intracelular de los virus a la membrana plasm\u00e1tica, bien sea por colas de actina o por otro mecanismo, depende de la adquisici\u00f3n de la envuelta. Se han descrito hasta el momento seis prote\u00ednas asociadas a al \u00faltima envuelta del eev. Los estudios recientes sobre estas prote\u00ednas se han centrado en su an\u00e1lisis bioqu\u00edmico y en el estudio de los mutantes de deleci\u00f3n.  en esta tesis se pretende dar un nuevo enfoque al estudio de la envuelta del eev, realizando la caracterizaci\u00f3n funcional de las prote\u00ednas implicadas, tanto en la infecci\u00f3n como expresadas individualmente en ausencia de infecci\u00f3n. las prote\u00ednas a incorporar en la envuelta han de estar presentes en el tgn en el momento en el que el virus adquiere la envuelta. As\u00ed, pues, durante la infecci\u00f3n, una o varias de estas prote\u00ednas han de quedar, de alguna manera retenidas en ese compartimento y determinar as\u00ed el sitio de envolvimiento del virus.  en primer lugar se estudi\u00f3 la localizaci\u00f3n celular de las prote\u00ednas de la \u00faltima envuelta del eev en una infecci\u00f3n comprobando que en c\u00e9lulas infectadas por vaccinia, las prote\u00ednas b5r, f13l y a56r mostraron patr\u00f3n de envuelta. Por el contrario, a56r se localiz\u00f3 en membrana plasm\u00e1tica, pero no produjo un patr\u00f3n atribuible a virus, a34r se localiz\u00f3 en un \u00e1rea yuxtanuclear de mayor extensi\u00f3n que el complejo de golgi y membrana plasm\u00e1tica.  en segundo lugar se estudi\u00f3 la presencia de se\u00f1ales de localizaci\u00f3n de las prote\u00ednas de la \u00faltima envuelta del eev mediante su expresi\u00f3n en ausencia de infecci\u00f3n utilizando el sistema de expresi\u00f3n basado en el semliki forest virus, comprobando que a34r y b5r conten\u00edan se\u00f1ales de localizaci\u00f3n intracelulares.
