Se emplearon sales de tetrazolio para monitorizar la viabilidad de los tejidos en cultivo. La tecnica de esterilizacion y la pared del ovario influyeron en la supervivencia de algunos tejidos del primordio seminal. Las plantulas, obtenidas por embriogenesis directa, fueron en su mayoria haploides. En la segunda parte se estudio la germinacion del polen de girasol in situ e in vitro. La tecnica de germinacion in vitro que mejores resultados dio fue la germinacion sobre un papel de celofan con un medio liquido compuesto por 400 g l-1 de sacarosa, 30 mg l-1 de acido borico y 600 mg l-1 de nitrato calcico. Con esta tecnica germino hasta un 50% de los granos de polen. En la ultima parte se combino la polinizacion con polen irradiado con rayos gamma y el cultivo in vitro de los embriones obtenidos. El polen irradiado (0-800 gy) germino normalmente tanto in vitro como in situ, pero perdio su capacidad de fecundacion con irradiaciones superiores a 100 gy. El polen de girasol puede ser considerado bastante sensible a la radiacion gamma.
