{"id":88810,"date":"2001-09-02T00:00:00","date_gmt":"2001-09-02T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/clonacion-y-regulacion-de-las-enzimas-del-complejo-cistea%c2%adna-sintasa-de-chlamydomonas-reinhardtii\/"},"modified":"2001-09-02T00:00:00","modified_gmt":"2001-09-02T00:00:00","slug":"clonacion-y-regulacion-de-las-enzimas-del-complejo-cistea%c2%adna-sintasa-de-chlamydomonas-reinhardtii","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/clonacion-y-regulacion-de-las-enzimas-del-complejo-cistea%c2%adna-sintasa-de-chlamydomonas-reinhardtii\/","title":{"rendered":"Clonacion y regulaci\u00f3n de las enzimas del complejo ciste\u00edna sintasa de chlamydomonas reinhardtii"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Cristina Gonz\u00e1lez Ravina <\/strong><\/h2>\n<p>La tesis aborda el estudio de la regulaci\u00f3n de las enximas o-acetilserina (tiol) liasa y serina acetiltranferasa de chlamydomonas reinhardtii, a fin de conocer mejor la ruta de asimilaci\u00f3n de azufre en dicho organismo.Utilizando diversas t\u00e9cnicas de biolog\u00eda molecular, se han aislado los genes que codifican dichas enzimas en este alga verde, y se han llevado a cabo estudios de actividdes enzim\u00e1ticas y nivel de expresi\u00f3n g\u00e9nica en dicho organismo bajo diferenes condiciones nutricionales de azufre y nitr\u00f3geno.  otro de los aspectos abordados en esta tesis ha sido la obtenci\u00f3n de anticuerpos policlonales frente a la enzima o-acetil serina (tiol) liasa de chlmydomonas, reinhardtil, con objeto de completar los estuidos de regulaci\u00f3n, y estudiar los niveles de dicha enzima a nivel de prote\u00edna en diferentes condiciones nutricionales as\u00ed como en c\u00e9lulas sometidas a limitaci\u00f3n de azufre.  todos los estudios realizados epara la estirpe silvestre de este alga verde, se llevaron tambi\u00e9n a cabo en dos estirpes mutantes sacl y sac3 de chlamydomonas, que se caracterizan por presentar una respuesta anormal a la limitaci\u00f3n nutricional. Los reultados obtendios sugieren un cierto acoplamiento regulatorio entre los mtabolismos de azufre y el nitr\u00f3geno, y adem\u00e1s muestra una coregulaci\u00f3n de ambas enzimas, serina acetiltransferasa y o-acetil serina (tiol) liasa en chalmydomonas reinhardtii.  chlamydomonas presenta adem\u00e1s una mayor capacidad para climatarse a la limitaci\u00f3n nutricional de azufre, siendo su respuesta r\u00e1pida casi a partir de la primera hora de tratamiento.  tambi\u00e9n se ha puesto a punto en esta tesis la determinaci\u00f3n de actividad enzim\u00e1tica sat en extractos
