Rosario G\u00f3mez Garc\u00eda <\/strong><\/h2>\n<p>Se ha realizado un estud\u00f1io sobre la pirofosfatasa inorg\u00e1nica solubre (sppasa, ec 3,6,1,1) de distintos organismo fotosint\u00e9ticos -cianobacterias, bacterias anoxig\u00e9nicas, microalgas y plantas- usando una aproximaci\u00f3n tanto bioqu\u00edmica como de gen\u00e9tica molecualr. Las sppasas hidrolizan la mol\u00e9cula de pirofosfato inorg\u00e1nico (ppi) en dos mol\u00e9culas de ortofosfato (pi) enuna reacci\u00f3n exerg\u00f3nica que permite que las reacciones biosint\u00e9ticas que liberan ppi trancurran en el sentido correcto. Se ha purifcado y caracterizado fisico-qu\u00edmicamente las sppasas de tres cianobacterias y una bacteria fotosint\u00e9tica anoxig\u00e9nica. Las sppasas de cianobacterias sonprote\u00ednas hexam\u00e9tricas (subunidad de 20 kda) mientras que las bacterias fotosinteticas presentanuna mayor diversidad de estados oligom\u00e9ricos. Se han clonado los genes ppa que codifican las sppasa cianobacterianas y se ha estudiado el efecto que produce la deficiencia en pi sobre la actividad sppasa de la cianobacteria synechocystis sp. Pcc 6803, as\u00ed como la inducci\u00f3n del transcrito del gen ppa en estas condiciones.Tambi\u00e9n se ha caracterizado las correspondientes sppasas de distitnas microalgas (chlanydomonas reinhardtti, cyanophora paradoxa, euglena fracilis, ochromonas danica) y de la planta spinacea oleracea, siendo en todos los casos prote\u00ednas de estructura monom\u00e9rica diferentes de sus hom\u00f3logas procariotas y localizadas en los org\u00e1nulos celualres (pl\u00e1stidos y moticondrias) ya que el citosol de los protists fotosint\u00e9ticos carece de actividad sppasa. de c. Reinhardtii se han purificado y caracterizado dos sppasas con diferente localizaci\u00f3ncelular denominadas sppasa 1 (cloropl\u00e1stica ) y sppasa 2 (mitocondrial). se ha identificado y validado por expresi\u00f3n heter\u00f3loga el cdna que codifica el polip\u00e9ptido precursos de la sppasa 1 de arabidopsis thaliana, que posee un p\u00e9ptido de tr\u00e1nsito n-terminal.
