Sin embargo, no se conocen los mecanismos por los que ejerce esta accion. Objetivos: 1) comprobar la proteccion por pge1 en la lha por d-galactosamina (d-ga1n). 2)estudiar el papel del oxido nitrico (no) sintetizado por el isoenzima inducible de la no-sintasa (inos) en este modelo y en la proteccion por pge1. 3) dilucidar el papel del tnfalfa en este modelo y en la proteccion por pge1. El trabajo se ha dividido en 3 bloques seg\u00fan los objetivos planteados. bloque 1. Metodos: 4 grupos de 20 ratas wistar (175-225 g): control, pge1 (250 ug\/kg), d-ga1n(1 g\/kg), pge1+d-ga1n. Cada grupo se subdividio en dos seg\u00fan fueran sacrificados a 4 y 12 h. La lha se valoro mediante el alt serico e histolog\u00eda. Resultados: pge1 disminuye la lha inducida por d-galn. bloque 2. Metodos: 8 grupos de 20 ratas: control, pge1, d-ga1n, pge1+d-galn, mt, mt+pge1, mt+d-galn, mt+pge1+d-galn. Cada grupo se subdividio en dos seg\u00fan fueran sacrificados a 4 y 12 h. La metil-isotiourea (mt) fue utilizada como inhibidor especifico de la inos, administrandose 2 dosis (5 mg\/kg) antes de la induccion de la lha y dos dosis adicionales a las 4 y 8 h. en el suero fueron determinados los niveles de alt, nitrito+nitrato y tnfalfa. en tejido hepatico se evaluo la lesion histologica y la expresion de inos. resultados: d-ga1n no incrementa los niveles de nitrito+nitrato en suero ni la expresion de inos en hepatocitos. Sin embargo, pge1 aumenta ambos parametros en el grupo pge1+d-ga1n. Bloque 3. Metodos: 8 grupos de 5 ratas: control, pge1, d-ga1n, pge1+d-ga1n. El anti-tnf-alfa (5 mg\/kg) se administro 1 h antes de los tratamientos. Los animales se sacrificaron a las 12h. se valoraron los parametros del bloque 2. Resultados: anti-tnfalfa+d-galn disminuye la lha por d-galn.
