{"id":89858,"date":"2018-03-10T00:15:09","date_gmt":"2018-03-10T00:15:09","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/el-paper-estructural-del-zinc-en-el-plegament-de-les-metal-lotioneines-de-coure\/"},"modified":"2018-03-10T00:15:09","modified_gmt":"2018-03-10T00:15:09","slug":"el-paper-estructural-del-zinc-en-el-plegament-de-les-metal-lotioneines-de-coure","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/el-paper-estructural-del-zinc-en-el-plegament-de-les-metal-lotioneines-de-coure\/","title":{"rendered":"El paper estructural del zinc en el plegament de les metal lotioneines de coure"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Roger Bofill Arasa <\/strong><\/h2>\n<p>Las metalotione\u00ednas mt son prote\u00ednas de bajo pedo molecular y elevado contenido en cys que se caracterizan por su extremada habilidad para enlazar cationes de metales pesados esenciales, como zn(ii) y cu(i), y t\u00f3xicos, como cd(ii), hg(ii) o ag(i).  este trabajo trata del papel estructural delzn(ii) en los procesos de plegamiento de mt de mam\u00edfero, insecto y crust\u00e1ceo en presencia de cu(i). las prote\u00ednas estudiadas han sido obtenidas mediante la t\u00e9cnica del adn recombiannte, la cual ha permitido obtenerlas con un elevado grado de pureza.  se han efecutado diferentes estudios sobre su capacidad coordinante in vitro e in vivo en presencia de cu(i) mediante el uso de diversas t\u00e9cnicas espectrosc\u00f3picas (uv-vis, dicroismo ciruclar (dc), emisi\u00f3n e icp \u00f3ptico) y mediante espectrometr\u00eda de masas con ionizaci\u00f3n por electrospray (esi-ms). los resultados obtenidos permiten concluir que el zn(ii) es indispensable para el plegamiento in vivo de la mt recombiante de rat\u00f3n y de la mt recombinante de bogavante americano (mth) en presencia de cu(i), mientras que en el caso de la mt recombinante de drosophila (mtn) el zn(ii) no presenta ning\u00fan papel.  adicionalmente, la afinidad de las mt frente a los metales fisiol\u00f3gicamente activos permite proponer un nuevo sistema de clasificaci\u00f3n de estas prote\u00ednas. as\u00ed, las mt que resultan en la formaci\u00f3n exclusiva de especies homomet\u00e1licas de cu(i) cuando son expresadas heter\u00f3logamente en un medio rico en cu s\u00f3n tion\u00ednas de cobre (p. Ej. Mtn), mientras que las mt que resultan en la formaci\u00f3n de especies heteromet\u00e1licas conteniendo a la vez cu(i) y zn(ii) son tione\u00ednas de zinc (p.Ej. Mt de rat\u00f3n, mth). Esta caracter\u00edstica se puede relacionar, mediante una an\u00e1lisis filogen\u00e9tica, con la estructura primaria de las mt. As\u00ed pues,la secuencia amonoac\u00edda de las mt es probablemente esencial para determinar la funcionalida de estas prote\u00ednas en los organismos vivos, la cual hoy en d\u00eda aun
