{"id":89920,"date":"2018-03-10T00:15:14","date_gmt":"2018-03-10T00:15:14","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/identificacio-de-nous-elements-reguladors-en-el-promotor-de-glut1\/"},"modified":"2018-03-10T00:15:14","modified_gmt":"2018-03-10T00:15:14","slug":"identificacio-de-nous-elements-reguladors-en-el-promotor-de-glut1","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/identificacio-de-nous-elements-reguladors-en-el-promotor-de-glut1\/","title":{"rendered":"Identificacio de nous elements reguladors en el promotor de glut1"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Manuela Sanchez Feutrie <\/strong><\/h2>\n<p>En esta tesis se han identificado nuevos elementos que dirigen la actividad transcripcional del promotor de gluti1 y su importancia en la regulaci\u00f3n de este promotor en un modelo de hipertrofia card\u00edaca y por la proteina supresora de tumores p53:  -se ha caracterizado una regi\u00f3n en la posici\u00f3n -46\/-37 del promotor de gluti1 de rata que une factores especificos de m\u00fasculo, de forma dependiente de diferenciaci\u00f3n y de desarrollo (mdde), de naturaleza desconocida. Mdde contribuye a la actividad basal del promotor tanto en mioblastos y miotubos como en cardiomitos neonatales y permite la m\u00e1xima estimulaci\u00f3n de la actividad transcripcional frente al ampc y sp1 en mioblastos l6e9, y al suero en cardiomiocitos neonatales. Hemos propuesto que mdde opera, tanto sin\u00e9rgicamente con sp1 como en ausencia de \u00e9ste, como lugar central en el mecanismo de regulaci\u00f3n del mantenimiento de la actividad transcripcional del promotor de glut1.  -otra regi\u00f3n estudiada ha sido la -69\/-55 en la que se ha determinado la uni\u00f3n de factores de transcripci\u00f3n de forma dependiente del estado de proliferaci\u00f3n y se requiere de la integridad de 8cs en el estado proliferativo (caja c8). La caja c8 regula negativamente la actividad transcripcional en c\u00e9lulas proliferativas de manera dependiente de suero.  -hemos identificado otra posible caja ap1(+41\/+62) que podr\u00eda ser, al menos en parte, responsable de los efectos estimuladores de c-jun.  -el tratamiento de cardiomiocitos neonatales con fenilefrina provoca un fenotipo hipertr\u00f3fico que provoca un incremento en la expresi\u00f3n de la prote\u00edna glut1. Una parte de este incremento de debe a una m\u00e1s elevada actividad transcripcional del promotor de glut1, y la integridad de las cajas gc, mdde,y tata es requerida en la respuesta. Adem\u00e1s las prote\u00ednas ras y c-jun est\u00e1n implicadas en estos efectos.  -la prote\u00edna p53 presentea efectos contrapuestos sobre la actividad transcripcional de glut1.
