La regi\u00f3n amino-terminal (ndp) de hsos1 est\u00e1 constituida por un dominio con homolog\u00eda a dbl (dh),un dominio con homolog\u00eda a pleckstrina (ph), y por una regi\u00f3n, que nosotros hemos denominado n-ter, sin homolog\u00eda conocida hasta hoy. La presente tesis doctoral es un estudio del efecto que los distintos dominios que componen esta regi\u00f3n amino-terminal (la regi\u00f3n n-ter, el dominio dh, el dominio ph y la regi\u00f3n dh-ph) tienen sobre la actividad de hsos1 y la funcionalidad en la activaci\u00f3n fisiol\u00f3gica de ras. Mediante ensayos de formaci\u00f3n de focos transformantes, actividad mapk y estado de activaci\u00f3n de ras, hemos podido determinar que la regi\u00f3n completa (ndp), as\u00ed como la regi\u00f3n dh-ph, tienen un papel regulador positivo mientras que la regi\u00f3n n-ter ejerce de regulador negativo; y el efecto de estos dominios sobre la activaci\u00f3n de ras es independiente del tipo de prote\u00edna ras (h, k o n-ras). Esta regi\u00f3n n-ter, correspondiente a los primeros 200 residuos de hsos1, ejerce dicha regulaci\u00f3n negativa mediante una interacci\u00f3n espec\u00edfica con el dominio ph, de tal modo que la regi\u00f3n dh-ph queda cubierta. Para identificar prote\u00ednas que puedan estar participando en estas regulaciones, por interacci\u00f3n con estos dominios, se ha realizado un ensayo de doble h\u00edbrido en levaduras. As\u00ed, hemos observado que la regi\u00f3n n-ter interacciona in vitro, in vivo y semi-in vivo (levaduras) con la prote\u00edna cgmp-pde9a (una fosfodiesterasa que hidroliza el segundo mansajero cgmp a su forma no ciclada) y con pa28a (subunidad reguladora del protesoma 26s). Esta regi\u00f3n dh-ph tambi\u00e9n interacciona semi-in vivo con las prote\u00ednas pa28b (otra subunidad reguladora del proteosoma 26s), p112 (reguladora del proteosoma 20s) y la caperonina
