Pilar Bayona Bafaluy <\/strong><\/h2>\n<p>Se conocen numerosas mutaciones en el mtdna que producen enfermedades en humanos. Es dif\u00edcil obtener modelos animales de estas enfermedades porque no se puede introducir dna ex\u00f3geno dentro de la mitocondria. Por esto, este trabajo describe una estrategia que permite aislar lineas celulares de rat\u00f3n con mutaciones en el mtdna. En esta estrategia se realiza en primer lugar una mutag\u00e9nesis al azar, a continuaci\u00f3n se manipula el n\u00c2\u00ba de copias de mtdna por c\u00e9lula de rat\u00f3n, utilizando un tratamiento que hemos probado que consiste en crecer clones celulares individuales en glucosa y en galactosa. los clones incapaces de vivir en galactosa posiblemente tendr\u00e1n da\u00f1ado su sistema oxphos. Para comprobarlo se mide su respiraci\u00f3n en electrodo de ox\u00edgeno.  para an\u00e1lisis de \u00e1bridos se ensaya si se trata de una mutaci\u00f3n mitocondrial o nuclear. Hemos aislado numerosos clones con mutaciones mitocondriales que producen defectos oxpmos que se transmiten con la mitocondria.
