Paz De La <\/strong><\/h2>\n<p>La interleuquina-6 (ik-6) es una citoquina que participa en los procesos inflamatorios desencadenados por el da\u00f1o tisular. La reparaci\u00f3n del tejido, subsecuente al da\u00f1o, incluye la remodelaci\u00f3n de la matriz extracelular (mec) en el \u00e1rea da\u00f1ada. Esta remodelaci\u00f3n supone la degradaci\u00f3n de los componentes de la matriz preexistente (por parte de las metalopretasas) y la s\u00edntesis de nuevas mol\u00e9culas. No exist\u00edan datos concluyentes sobre el papel de la il-6 en la remodelaci\u00f3n de la mec. En este trabajo se analiz\u00f3 la regulaci\u00f3n de una metaloproteasa, la colagenasa intersticial, por la il-6 en cultivos de fibroblastos de rata de la l\u00ednea rat2.  la il-6 (20 ng\/ml) aument\u00f3 la expresi\u00f3n del gen de la colagenasa intersticial de rata a partir de 6 horas de tratamiento y el m\u00e1ximo se alcanz\u00f3 a las 24 horas. Igualmente la il-6 aument\u00f3 la producci\u00f3n de colagenasa, de 60 kda, y su secreci\u00f3n a los medios de cultivo. En paralelo al aumento de la expresi\u00f3n del gen hallamos que la il-6 induce la actividad de un fragmento del promotor de la colagenasa de 2000 pares de bases. El an\u00e1lisis mutacional mostr\u00f3 que el sitio ap-1, situado entre las posiciones -19 y -42 en este promotor, determina la respuesta a la il-6. La il-6 aument\u00f3 el grado de uni\u00f3n del factor ap-1 a su secuencia consenso. Cuando se analiz\u00f3 el papel de la il-6 en la regulaci\u00f3n de las prote\u00ednas que forman el factor ap-1 hallamos que la il-6 induce los arnm de c-jun y c-fos, la actividad de sus promotores y los niveles intracelulares de estas dos prote\u00ednas. Asimismo el tratamiento con la il-6 supuso un aumento de los niveles de c-jun fosforilado en la serina-63. Esta fosforilaci\u00f3n determina la actividad del factor ap-1 y est\u00e1 mediada por la quinasa n-terminal de jun (jnk). Parad\u00f3jicamente la il-6 no indujo la actividad jnk.
