Esta quimasa desempe\u00f1a un papel muy importante en la desensibilizaci\u00f3n y se\u00f1alizaci\u00f3n de numerosos receptores, y sus niveles y funcionalidad se encuentran alterados en diversas patolog\u00edas como hipertensi\u00f3n, hipotiroidismo o fallo cardiaco congestivo. El trabajo realizado en esta tesis ha permitido proponer un mecanismo funcional para la activaci\u00f3n y translocaci\u00f3n de grk2 en respuesta a est\u00edmulo basado en la modulaci\u00f3n de la actividad de grk2 frente a distintos sustratos por dominio n y c-terminales de la quinasa, junto con el efecto protector de un p\u00e9ptido c-terminal en su proteolisis parcial, y en la elaboraci\u00f3n de un modelo molecular para grk2. En este modelo, grk2 se mantendr\u00eda en un estado basal inactivo mediante interacciones intramoleculares entre los dominios rgs, ph y catal\u00edtico. La ruptura de esos contactos por interacci\u00f3n con subunidades gbetay, l\u00edpidos y receptores activados promover\u00edan cambios conformacionales que simult\u00e1neamente translocan grk2 a la membrana y la activan. Se ha demostrado tambi\u00e9n la regulaci\u00f3n de la quinasa por fosforilaci\u00f3n por otras quinasas activadas por receptores acoplados a prote\u00ednas g como src y mapk. Grk2 recombinante se fosforila por la tirosina quinasa c-src con rapidez, alta afinidad, y estequiom\u00e9tricamente.La fosforilaci\u00f3n de grk2 en residuos de tirosina tienen lugar tambi\u00e9n en un contexto celular, tanto en sistemas heter\u00f3logos, como end\u00f3genos, en respuesta a la estimulaci\u00f3n de diversos receptores acoplados a distintas prote\u00ednas g, entre  los que se encuentran los receptores beta2- y alfa2a-adren\u00e9rgicos, el receptor m1-muscar\u00ednico de acetilcolina, y el receptor de quimioquinas cxcr4. Por tanto, este mecanismo de regulaci\u00f3n de grk2 por fosforilaci\u00f3n ser\u00eda inherente a la estimulaci\u00f3n de m\u00faltiples receptores acoplados a prote\u00ednas g.
