{"id":91291,"date":"2018-03-11T10:09:58","date_gmt":"2018-03-11T10:09:58","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/mecanisme-enzima-tic-de-la-13-14-a-glucanasa-de-bacillus-licheniformis-estudis-cina%c2%a8tics-en-estat-estacionari-i-preestacionari\/"},"modified":"2018-03-11T10:09:58","modified_gmt":"2018-03-11T10:09:58","slug":"mecanisme-enzima-tic-de-la-13-14-a-glucanasa-de-bacillus-licheniformis-estudis-cina%c2%a8tics-en-estat-estacionari-i-preestacionari","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/bioquimica\/mecanisme-enzima-tic-de-la-13-14-a-glucanasa-de-bacillus-licheniformis-estudis-cina%c2%a8tics-en-estat-estacionari-i-preestacionari\/","title":{"rendered":"Mecanisme enzim\u00c1\u00a0tic de la 1,3-1,4-\u00c2\u00bf-glucanasa de bacillus licheniformis: estudis cin\u00e9tics en estat estacionari i preestacionari"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Mireia Abel Lluch <\/strong><\/h2>\n<p>Las glicosidasas que hidrolizan substratos con retenci\u00f3n de configuraci\u00f3n siguen un mecanismo general en que despu\u00e9s de una primera etapa de asociaci\u00f3n enzima-ligando se encadenan dos etapas catal\u00edticas que llevan a la hidr\u00f3lisis del substrato conservando la configuraci\u00f3n del carbono anom\u00e9rico en el nuevo extremo generado.     en el presente trabajo se comprueba a trav\u00e9s de estudios mecan\u00edsticos, principalmente estudios en estado pre-estacionario y an\u00e1lisis de hammett, que con este mecanismo tan sencillo no se puede describir correctamente la actividad catal\u00edtica de la 1,3-1,4-\u00edY-glucanasa de bacillus licheniformis, y se propone el mecanismo que se muestra a continuaci\u00f3n:     donde los complejos enzima-substrato adoptan dos conformaciones productivas (ses* y ses**) y donde el balance entre estas dos conformaciones en funci\u00f3n del substrato explica los diferentes comportamientos cin\u00e9ticos en estado estacionario y pre-estacionario. un estudio en profundidad sobre la importancia que ejerce la interacci\u00f3n entre el enzima y el hidroxilo en c2 de la unidad de glucopiranosa que ocupa el subsitio -1 demuestra que en el caso de la 1,3-1,4-\u00edY-glucanasa de bacillus licheniformis esta interacci\u00f3n no juega el papel relevante que se ha observado en otras \u00edY-glicosidasas que actuan con retenci\u00f3n de configuraci\u00f3n. el estudio de la reacci\u00f3n de hidrataci\u00f3n del glical g4g3g catalizada por la 1,3-1,4-\u00edY-glucanasa de bacillus licheniformis permite proponer que hay un cambio en el residu que ejerce el papel de \u00e1cido general. De manera que se propone que el asp136 que en la reacci\u00f3n de hidr\u00f3lisis de substratos ayuda en el posicionamiento de los res\u00edduos catal\u00edticos y juega un papel en la regulaci\u00f3n de sus pka, es el res\u00edduo que act\u00faa de \u00e1cido general en la reacci\u00f3n de hidrataci\u00f3n de glicales.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
