Mediante organog\u00e9nesis adventicia es una herramienta biotecnol\u00f3gica que permite la obtenci\u00f3n de individuos clonales as\u00ed como establecer un sistema experimental adecuado para el estudio de las bases fisiol\u00f3gicas del proceso caulog\u00e9nico. El objetivo de la tesis doctoral ha sido la optimizaci\u00f3n del protocolo de micropropagaci\u00f3n de 6 familias seleccionadas, evaluando su respuesta organog\u00e9nica y certificando la estabilidad gen\u00e9tica de los individuos obtenidos mediante distintos marcadores moleculares (rapd, aflp, issr y sampl), resultados que tambi\u00e9n apoyan la baja variabilidad gen\u00e9tica descrita para esta especie. Adem\u00e1s, ensayos de transferencia de medios e histol\u00f3gicos han permitido establecer etapas discretas durante el periodo de inducci\u00f3n caulog\u00e9nica. Complementando estos resultados, an\u00e1lisis hormonales tanto de absorci\u00f3n y metabolismo del agente inductor (la citoquinina benciladenina) como de niveles end\u00f3genos de citoquininas tanto arom\u00e1ticas como isopreno\u00eddicas han permitido definir las din\u00e1micas asociadas a la inducci\u00f3n de yemas en cotiledones maduros de p. Pinea, confirmando como naturales, por primera vez en con\u00edferas, las citoquininas arom\u00e1ticas benciladenina, rib\u00f3sido de benciladenina y para-topolina. Tras seleccionar las familias que presentaban una mayor y menor intensidad de respuesta caulog\u00e9nica, se ha podido asociar una mayor absorci\u00f3n de la benciladenina aplicada ex\u00f3genamente al medio de cultivo con una mayor respuesta caulog\u00e9nica. Tambi\u00e9n en las primeras etapas de cultivo existe una relaci\u00f3n entre mayor magnitud de respuesta caulog\u00e9nica y mayores niveles end\u00f3genos de benciladenina, rib\u00f3sido de benciladenina y meta-topolina, mientras que la menor intensidad de respuesta caulog\u00e9nica se asocia con mayores niveles end\u00f3genos de rib\u00f3tidos y gluc\u00f3sidos arom\u00e1ticos.  the micropropagation of pinus pinea l. Families by adventitious organogenesis is a suitable biotechnological tool for not only obtaining clonal individuals but also for setting an experimental system for going depth into the physiological bases of the caulogenic process. The main objective of the doctoral thesis has been the optimization of a micropropagation procedure for 6 selected half-sibling families. It has also been evaluated their organogenic response, certifying the genetic stability by using different molecular markers (rapd, aflp, issr and sampl), these results also support the low genetic variability proposed to this species. Additionally, media transfer assays and histological approaches have allowed us the establishment of several discrete phases during the caulogenic induction. Furthermore, hormonal analyses of absorption and metabolism of the inductor agent (the cytokinin benzyladenine) and other natural occurring cytokinins, both aromatic and isoprenoidic type, have defined the dynamics associated with the bud induction on mature cotyledons of p. Pinea. For the first time in conifers, the aromatic cytokinins benzyladenine, benzyladenine riboside, and para-topolin are confirmed as naturally occurring cytokinins. After selecting those families with extremely intensity of response, it has been matched that more uptake of exogenously applied benzyladenine leads to more caulogenic response intensity. During the first hours of induction culture, there is a relationship between a high magnitude of response and higher levels of benzyladenine, benzyladenine riboside and meta-topolin.
