{"id":92957,"date":"2018-03-11T10:12:08","date_gmt":"2018-03-11T10:12:08","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/mecanismo-de-deplecion-de-glutation-y-papel-de-la-sera%c2%adn-treona%c2%adn-fosfatasa-pp2a-en-la-pancreatitis-aguda\/"},"modified":"2018-03-11T10:12:08","modified_gmt":"2018-03-11T10:12:08","slug":"mecanismo-de-deplecion-de-glutation-y-papel-de-la-sera%c2%adn-treona%c2%adn-fosfatasa-pp2a-en-la-pancreatitis-aguda","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/bioquimica-molecular\/mecanismo-de-deplecion-de-glutation-y-papel-de-la-sera%c2%adn-treona%c2%adn-fosfatasa-pp2a-en-la-pancreatitis-aguda\/","title":{"rendered":"Mecanismo de depleci\u00f3n de glutati\u00f3n y papel de la ser\u00edn\/treon\u00edn fosfatasa pp2a en la pancreatitis aguda"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Justo Javier Escobar Cubiella <\/strong><\/h2>\n<p>En la presente tesis doctoral se han estudiado los mecanismos implicados en la degradaci\u00f3n de gsh en los modelos in vivo de pa necr\u00f3tica inducida por infusi\u00f3n retr\u00f3grada de taurocolato s\u00f3dico al 3,5% y de pa edematosa inducida por una dosis supram\u00e1xima de cerule\u00edna, confirmando que la depleci\u00f3n mantenida de gsh favorecen el desarrollo de la pa necr\u00f3tica y que a pesar de que el mecanismo de s\u00edntesis de la g-glutamil ligasa (gcl), mediado por erk 1\/2 y el factor de transcripci\u00f3n c-myc, aparece activo tanto en la pa edematosa como en la necr\u00f3tica, en la necr\u00f3tica se produce un fallo del mecanismo  originado por la activaci\u00f3n de la ribonucleasa (rnasa) pancre\u00e1tica en el citosol de las c\u00e9lulas acinares responsable de la degradaci\u00f3n del arnm de la gcl.  adem\u00e1s se ha caracterizado la tasa de degradaci\u00f3n de gsh y de formaci\u00f3n de glutamato y ciste\u00edna en extractos citos\u00f3licos de p\u00e1ncreas de rata y la inhibici\u00f3n de estas tasas por el inhibidor de ser\u00edn peptidasas amino etil bencenil sulfonil fluoruro (aebsf) en el modelo in vivo de pa necr\u00f3tica.  Tambi\u00e9n se ha realizado cromatograf\u00eda de afinidad con columnas de retenci\u00f3n de gsh (1) y de aebsf-biotinilado (2) en extractos citos\u00f3licos de ratas con pa identificando prote\u00ednas gsh transferasas y glutarredoxina (1) o  htra 3a, rnasa pancre\u00e1tica, tripsina, arid 4d (rbpl 1) y arginina: glicina amidino transferasa (2). Tambi\u00e9n se ha descartado a la tripsina,  gamma-glutamil transpeptidasa y carboxipeptidasa b pancre\u00e1tica como posibles efectoras directas de la degradaci\u00f3n del gsh. en cuanto a la cascada inflamatoria se ha determinado que durante la pa se produce la disminuci\u00f3n de las ser\u00edn treon\u00edn fosfatasas pp2a, pp2b (calcineurina) y pp2c 1 hora despu\u00e9s de la inducci\u00f3n de la pa necr\u00f3tica y de las tiros\u00edn fosfatasas citos\u00f3licas que podr\u00eda favorecer el aumento de la fosforilaci\u00f3n de las mapk en la pa. en estudios in vitro, se ha estudiado la actividad de la pp2a en c\u00e9lulas ar42j incubadas con taurocolato al 0,3% produci\u00e9ndose una disminuci\u00f3n transitoria de su actividad indicando la implicaci\u00f3n de la pp2a en fen\u00f3menos de se\u00f1alizaci\u00f3n celular. Adem\u00e1s la ptx es capaz de prevenir esa perdida de la actividad pp2a mediado por el ampc.   por inmunoprecipitaci\u00f3n de la cromatina (chip) se ha visto que durante la pa se produce el reclutamiento de acetiltransferasas a los promotores de genes inflamatorios como egr-1, inos-2, icam-1 y tnf-a y  la ptx es capaz de bloquear la uni\u00f3n de las acetiltransferasas cbp y  promover la
