Se estudia la modulaci\u00f3n de las cinasas de tirosina (tyrk) sobre la viabilidad celular (con el test mtt), la liberaci\u00f3n de histamina (con el met\u00f3do de shore), el ca2+ citos\u00f3lico (con fura-2) y el ph intracelular (con bcecf). Se utilizan los inhibidores de tyrk sti571, lavendustina a y genisteina en las dos subl\u00edneas hmc-1 y se observa que el f\u00e1rmaco sti571 reduce la liberaci\u00f3n espont\u00e1nea de histamina en c\u00e9lulas hmc-1560 y hmc-1560,816 a trav\u00e9s de tyrk diferentes de las que se activan por el receptor c-kit. Adem\u00e1s sti571 aumenta la liberaci\u00f3n de histamina inducida por una alcalinizaci\u00f3n en c\u00e9lulas hmc-1560 y hmc-1560,816. dado que el ph intracelular tiene una gran importancia en las c\u00e9lulas hmc-1, se incuban las c\u00e9lulas en medios sin hco3- o sin na+\/hco3-. De esta forma se definen los intercambiadores implicados en la regulaci\u00f3n del phi basal. Adem\u00e1s se determina como estos transportadores afectan a la viabilidad celular y a la liberaci\u00f3n de histamina inducida por una alcalinizaci\u00f3n. De esta forma se concluye que las c\u00e9lulas hmc-1560 y hmc-1560,816 expresan el intercambiador nhe1 y un transportador de hco3- dependiente de na+ que es m\u00e1s sensible en las c\u00e9lulas con dos mutaciones. Adem\u00e1s el transportador de hco3- y la presencia de este i\u00f3n en el medio extracelular son imprescindibles para la supervivencia de las c\u00e9lulas hmc-1560 y hmc-1560,816 y participan en la alcalinizaci\u00f3n citos\u00f3lica en las dos subl\u00edneas.  se ha observado que la pkc modula la desgranulaci\u00f3n de mastocitos. En este contexto se estudian los efectos de la estimulaci\u00f3n de pkc sobre la liberaci\u00f3n de histamina, los niveles de ca2+ citos\u00f3lico y la expresi\u00f3n de c-kit (cit\u00f3metro de flujo). Se observa que en las c\u00e9lulas hmc-1560 y hmc-1560,816, la pkc delta es un regulador negativo de la liberaci\u00f3n de histamina inducida por un ion\u00f3foro de ca2+. Adem\u00e1s la activaci\u00f3n de la pkc o un aumento en el ca2+ citos\u00f3lico reduce por distintos mecanismos la expresi\u00f3n del receptor c-kit en las c\u00e9lulas hmc-1560 y hmc-1560,816.
