Se origina como resultado de la transformaci\u00f3n maligna de las c\u00e9lulas epiteliales que recubren el interior del colon y recto. Estudios epidemiol\u00f3gicos y precl\u00ednicos indican que la 1,25-dihidroxivitamina d3 (1,25(oh)2d3), el metabolito m\u00e1s activo de la vitamina d en nuestro organismo, tiene efectos protectores frente al crc. Nuestro laboratorio ha demostrado que la 1,25(oh)2d3 y algunos de sus an\u00e1logos inhiben la proliferaci\u00f3n y promueven la diferenciaci\u00f3n epitelial de c\u00e9lulas (sw480-adh y otras) de c\u00e1ncer de colon. La 1,25(oh)2d3 altera dr\u00e1sticamente el perfil de expresi\u00f3n g\u00e9nica celular, con inducci\u00f3n transcripcional del gen cdh1\/e-cadherina e inhibici\u00f3n de la ruta wnt\/\u00edY-catenina, muy frecuentemente activada en esta neoplasia.  los resultados presentes en esta memoria muestran que la expresi\u00f3n de sprouty-2 (spry2, una prote\u00edna moduladora de la se\u00f1alizaci\u00f3n mediada por receptores tirosina quinasa) es inhibida por la 1,25(oh)2d3. Esta inhibici\u00f3n tiene lugar a trav\u00e9s de mecanismos dependientes e independientes de e-cadherina\/adhesi\u00f3n celular. A su vez, la expresi\u00f3n ect\u00f3pica de spry2 en c\u00e9lulas sw480-adh inhibe los niveles de expresi\u00f3n de e-cadherina, tanto basales como inducidos por 1,25(oh)2d3. Como consecuencia, spry2 impide la inducci\u00f3n por la 1,25(oh)2d3 del fenotipo adhesivo. El estudio en detalle de esta represi\u00f3n mutua entre spry2 y e-cadherina revel\u00f3 que sus respectivos niveles de expresi\u00f3n se correlacionan inversamente en l\u00edneas celulares de c\u00e1ncer de colon y en tumores generados por c\u00e9lulas humanas transplantadas en ratones inmunodeficientes. El an\u00e1lisis de tumores de colon humanos confirm\u00f3 esta relaci\u00f3n inversa y mostr\u00f3 un aumento de los niveles de spry2 en tumores moderada y pobremente diferenciados respecto a tumores bien diferenciados, que expresan altos niveles de e-cadherina. Adem\u00e1s la expresi\u00f3n ect\u00f3pica de spry2 impide otras acciones de la 1,25(oh)2d3, como el antagonismo en la ruta wnt\/beta-catenina y la inhibici\u00f3n de la proliferaci\u00f3n celular. finalmente, hemos comprobado que spry2 reprime tambi\u00e9n la expresi\u00f3n de claudina-7 y ocludina, prote\u00ednas de las uniones ocluyentes, y que los niveles de claudina-7 en tumores de colon
