En humanos, nkg2d reconoce a dos familias de prote\u00ednas denominadas mica\/b y ulbps cuya expresi\u00f3n se induce como consecuencia de procesos de estr\u00e9s como la transformaci\u00f3n tumoral. La uni\u00f3n de este receptor y sus ligandos desencadena respuestas citot\u00f3xicas que pueden causar la lisis de las c\u00e9lulas transformadas o infectadas por virus. El objetivo principal de esta tesis ha sido estudiar los mecanismos moleculares involucrados en la regulaci\u00f3n de la expresi\u00f3n de los ligandos de nkg2d en c\u00e9lulas tumorales, tanto aquellos que permiten la transcripci\u00f3n basal de estos genes, como los fen\u00f3menos que reprimen su expresi\u00f3n con el fin de evadir la inmunovigilancia del c\u00e1ncer. As\u00ed, hemos demostrado que los factores de transcripci\u00f3n sp1 y, principalmente, sp3 son esenciales para la expresi\u00f3n basal de ulbp1 en c\u00e9lulas tumorales. Adem\u00e1s, nuestros experimentos revelaron que el factor ap-2\u00c2\u00bf es un represor de la expresi\u00f3n de este gen en c\u00e9lulas hela. Por otro lado, hemos demostrado que la deacetilasa de histonas 3 (hdac3) ejerce un papel represor de la transcripci\u00f3n de las ulbps en l\u00edneas celulares epiteliales tumorales. De forma coherente, el tratamiento con la tricostatina a (tsa), un inhibidor de las hdacs, induce la expresi\u00f3n de las ulbps y, en conescuencia, el reconocimiento y la lisis de c\u00e9lulas tumorales por parte de c\u00e9lulas nk. Finalmente, nuestros estudios revelan que la expresi\u00f3n de los ligandos de nkg2d es incrementada en el proceso de transici\u00f3n epitelio-mes\u00e9nquima (emt), causada por el tratamiento con el
