{"id":94956,"date":"2009-08-07T00:00:00","date_gmt":"2009-08-07T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-del-sistema-inmune-innato-y-adaptativo-en-urta-pagrus-auriga\/"},"modified":"2009-08-07T00:00:00","modified_gmt":"2009-08-07T00:00:00","slug":"estudio-del-sistema-inmune-innato-y-adaptativo-en-urta-pagrus-auriga","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/cadiz\/estudio-del-sistema-inmune-innato-y-adaptativo-en-urta-pagrus-auriga\/","title":{"rendered":"Estudio del sistema inmune innato y adaptativo en urta (pagrus auriga)"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Araceli Beatriz Mart\u00edn Antonio <\/strong><\/h2>\n<p>En esta tesis se estudia el sistema inmune de la urta (pagrus auriga), dividiendo su estudio en dos partes, el sistema inmune innato (sii) y en el sistema inmune adaptativo (sia). para el estudio del sii, mediante la t\u00e9cnica de hibridaci\u00f3n sustractiva supresiva (ssh) se obtuvieron tras inducci\u00f3n con lps (est\u00edmulo bacteriano) ests correspondientes a genes expresados diferencialmente debido a este tratamiento. De estos genes obtenidos, se llev\u00f3 a cabo una selecci\u00f3n de 4 relacionados con la respuesta del sii para obtener sus secuencias completas, determinando los elementos conservados y caracter\u00edsticos de cada familia y se procedi\u00f3 a estudiar mediante pcr a tiempo real su expresi\u00f3n durante la ontogenia de p. Auriga, su expresi\u00f3n en los distintos \u00f3rganos de individuos juveniles y su inducci\u00f3n tras tratamiento con lps y la bacteria photobacterium damselae subsp damselae en ri\u00f1\u00f3n cef\u00e1lico analizando la variaci\u00f3n temporal de la inducci\u00f3n desde la hora a las 48 horas tras el est\u00edmulo. En un experimento similar se estudi\u00f3 igualmente la inducci\u00f3n tras lps a las 3 horas en ri\u00f1\u00f3n cef\u00e1lico, h\u00edgado y bazo. Los genes seleccionados para este estudio fueron: la hepcidina, una quimioquina cxc, el factor inhibidor de la migraci\u00f3n de macr\u00f3fagos (mif), y el factor regulador de interfer\u00f3n 1 (irf-1).  respecto al estudio del sia, se llev\u00f3 a cabo la producci\u00f3n de un anticuerpo policlonal contra ig de p. Auriga para su uso en inmunoensayos, para ello se obtuvo y caracteriz\u00f3 la igm de p. Auriga, y mediante western blot y elisa se mostr\u00f3 la reactividad y especificidad del antisuero obtenido en suero de urta e ig purificada. Mediante citometr\u00eda de flujo se determinaron las poblaciones leucocitarias reconocidas por el antisuero en ri\u00f1\u00f3n cef\u00e1lico, mostr\u00e1ndose tambi\u00e9n en otros tejidos inmunes (bazo, intestino, branquias y ri\u00f1\u00f3n cefalico) el % de c\u00e9lulas portadoras de ig que reconoc\u00eda el antisuero. Se determin\u00f3 mediante western blot y elisa la reactividad cruzada del antisuero en otras especies de peces y por \u00faltimo se mostr\u00f3 la aplicabilidad del antisuero obtenido para ver la respuesta tras vacunaci\u00f3n en individuos p.
