Garcia Rubio <\/strong><\/h2>\n<p>La regulaci\u00f3n de la expresi\u00f3n g\u00e9nica responde fundamentalmente a una necesidad funcional de las c\u00e9lulas. Existen elementos que tienen un efecto importante sobre la transcripci\u00f3n que incluyen desde los factores generales de la transcripci\u00f3n, que act\u00faan esencialmente sobre la iniciaci\u00f3n, a elementos estructurales del adn. Entre estos destacan la estructura de la cromatina y el superenrollamiento local del adn. El superenrollamiento se origina cuando se giran las hebras en torno al eje de la doble h\u00e9lice, para perder o ganar torsi\u00f3n, y se unen de nuevo los extremos. En las c\u00e9lulas, existen enzimas que catalizan precisamente este proceso. Se las denomina topoisomerasas, pues convierten unos topois\u00f3meros en otros. Hay topoisomerasas de tipo i, las cuales pueden relajar tanto superenrollamiento negativo como positivo, act\u00faan sobre el adn monocatenario. Otras topoisomerasas, denominadas de tipo ii, son  esenciales para la replicaci\u00f3n y transcripci\u00f3n del adn, as\u00ed como para la recombinaci\u00f3n y segregaci\u00f3n cromos\u00f3mica. Esta enzima hace posible el acceso al adn mediante rotura y reparaci\u00f3n de la doble cadena. Es la prote\u00edna hom\u00f3loga a la adn girasa bacteriana. partimos de la base de que la topolog\u00eda del adn debe ser un factor determinante de la eficiencia de la elongaci\u00f3n, ya que el avance de la polimerasa  de arn (rnap) produce una acumulaci\u00f3n de superenrollamiento positivo por delante. Aunque hay bastante informaci\u00f3n del efecto de la estructura de la cromatina en iniciaci\u00f3n de la transcripci\u00f3n in vivo, poca informaci\u00f3n existe sobre el requerimiento topol\u00f3gico del adn para la transcripci\u00f3n, en particular durante la elongaci\u00f3n. Sin embargo, el hecho de que en su avance, la polimerasa ii de arn elongante elimine superenrollamiento positivo por delante y acumule negativo por detr\u00e1s, hace necesaria una acci\u00f3n coordinada de las topoisomerasas  como ya puso de manifiesto el trabajo de r. Sternglanz en los a\u00f1os 80 en la levadura s.
