Una vez redise\u00f1ados se han sintetizado mediante la t\u00e9cnica de pcr y se han introducido en el vector de expresi\u00f3n ppiczalphaa, t\u00edpico de pichia y para la expresi\u00f3n controlada por el promotor de la alcohol oxidasa (aox). se han obtenido 4 tipos distintos de cepas, unas que pueden expresar el gen alpha, otras el beta y las \u00faltimas el heterod\u00edmero, introduciendo los genes en los dos \u00f3rdenes posibles. se ha analizado la capacidad productiva preliminar de estas cepas mediante cultivos en erlenmeyer. Seguidamente se han hecho cultivos en batch de 1 y 3l, as\u00ed como en fed-batch de 3 y 5l. el caldo de cultivo se ha usado para implementar el proceso de purificaci\u00f3n. Una purificaci\u00f3n a partir de la cola de histidinas no ha sido posible y se ha sustituido por dos etapas de cromatograf\u00eda, una de intercambio i\u00f3nico y otra de gel-filtraci\u00f3n. una vez obtenida la prote\u00edna recombinante purificada se ha procedido en una breve caracterizaci\u00f3n bioqu\u00edmica, como son la determinaci\u00f3n del peso molecular, punto isoel\u00e9ctrico y el contenido en az\u00facares. Se ha comparado con la hormona nativa. Tambi\u00e9n se ha analizado su actividad in vitro y in vivo, dando positiva la primera y negativa la segunda. As\u00ed pues, hace falta reformular la hormona para poder utilizarla en reproducci\u00f3n animal y que sea activa cuando se administra.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
