En este trabajo, hemos estudiado la contribuci\u00f3n de la liberaci\u00f3n de ca2+ por parte del re a trav\u00e9s de los receptores de rianodina (ryr) e ip3 (ip3r) a la excitotoxicidad en modelos neuronales y oligodendrogliales in vitro. Una vez comprobada la expresi\u00f3n de los ryrs e ip3rs en los modelos celulares utilizados, se realizaron ensayos de excitotoxicidad en presencia de inhibidores de la liberaci\u00f3n de ca2+ a trav\u00e9s de los receptores mencionados anteriormente. Los est\u00edmulos con nmda y ampa produjeron una muerte celular dosisdependiente en neuronas y oligodendrocitos respectivamente, que se vio significativamente reducida en presencia de inhibidores de la liberaci\u00f3n de ca2+ a trav\u00e9s del re. Adem\u00e1s, la rianodina (inhibidora de los ryr) y el 2apb (bloqueante de la v\u00eda de los ip3r) disminu\u00edan el incremento de ca2+ citos\u00f3lico y la generaci\u00f3n de especies reactivas de ox\u00edgeno inducidas por el est\u00edmulo excitot\u00f3xico, mientras que el 2apb redujo la despolarizaci\u00f3n de la membrana mitocondrial y la activaci\u00f3n de la caspasa-3. De acuerdo con una alteraci\u00f3n de la homeostasis del re, observamos un estr\u00e9s de re inducido por nmda y ampa en neuronas y oligodendrocitos respectivamente, caracterizado por una fosforilaci\u00f3n de eif2\u00c2\u00bf y una sobreexpresi\u00f3n de chaperonas, fen\u00f3menos que se vieron regulados por inhibidores de la liberaci\u00f3n de ca2+ desde el re. en conjunto, estos resultados demuestran que la liberaci\u00f3n de ca2+ desde el re a trav\u00e9s de los ryrs e ip3rs contribuye a la muerte neuronal y oligodendroglial, promoviendo la disfunci\u00f3n mitocondrial e
