En este trabajo se han estudiado aspectos diferentes que participan en los procesos de homeostasis de cationes en levaduras.  utilizando como modelo la levadura saccharomyces cerevisiae, hemos caracterizado el transporte de potasio en una cepa silvestre y en una cepa doble mutante trk1 trk2, carente de los principales sistemas de transporte de potasio. Esta parte de la tesis se ha realizado en el contexto de un proyecto de biolog\u00eda de sistemas, y uno de los primeros pasos para abordarlo fue el dise\u00f1o de un medio de cultivo estandarizado, con concentraciones de k+ y na+ muy bajas (15 mm y 1,3 mm respectivamente).   En estas condiciones, se analizaron las caracter\u00edsticas del transporte, la concentraci\u00f3n interna del cati\u00f3n y el volumen celular.  en este trabajo tambi\u00e9n hemos identificado, clonado y expresado el gen ppz1 de la levadura debaryomyces hansenii, una levadura halotolerante que ofrece ventajas para la identificaci\u00f3n de nuevos factores gen\u00e9ticos implicados en los mecanismos de resistencia a sal. Al expresar el gen dhppz1 en una cepa de s. Cerevisiae mutante en los genes ppz1 y ppz2, se observ\u00f3 como la cepa que expresaba dhppz1 mostraba una mayor sensibilidad a na+, li+, tetrametilamonio e higromicina b, a la vez que exhib\u00eda una mayor tolerancia a cafe\u00edna. Adem\u00e1s, estas c\u00e9lulas presentaban un menor contenido de k+ respecto al control y por tanto un volumen celular tambi\u00e9n inferior.  finalmente, hemos completado un primer estudio sobre el estr\u00e9s oxidativo en d. Hansenii y se muestra por primera vez la naturaleza sensible de d.
