Pancreatitis y c\u00e1ncer), el estudio de las modificaciones producidas durante la diferenciaci\u00f3n acinar en dichas patolog\u00edas, se ha visto limitado por la escasez de modelos celulares no tumorales. Resultados previos de nuestro laboratorio, muestran que las c\u00e9lulas mes (c\u00e9lulas madre embrionarias de rat\u00f3n) pluripotentes y con la capacidad para generar tipos celulares especializados- pueden desarrollar un fenotipo acinar in vitro.  Los objetivos de esta tesis han sido aumentar el contenido de enzimas digestivos as\u00ed como las propiedades funcionales de las c\u00e9lulas generadas. Para ello se sobreexpresaron de forma estable  p48, rbpjl y mist1en c\u00e9lulas madre por transducci\u00f3n lentiviral de estos genes.  obtuvimos, gracias a una estrategia de infecci\u00f3n en m\u00faltiples etapas, l\u00edneas celulares transg\u00e9nicas mes que expresaban de forma constitutiva rbpjl y\/o mist1.La superimposici\u00f3n de la expresi\u00f3n de p48 por infecci\u00f3n lentiviral en c\u00e9lulas en proceso de diferenciaci\u00f3n dio lugar a una fuerte expresi\u00f3n de enzimas digestivos, con un patr\u00f3n de regulaci\u00f3n similar al que acontece in vivo durante el desarrollo pancre\u00e1tico. En esta inducci\u00f3n, tanto p48 como rpbjl son indispensables. Por otro lado, hemos mostrado un aumento elevado en la producci\u00f3n de varios componentes de la maquinaria secretota dependiente de mist1. Adem\u00e1s, hay que hacer notar, que las c\u00e9lulas p48\/rbpjl\/mist1 exhiben una regulada-secreci\u00f3n en respuesta a los secretagogos acinares y una mejor actividad de que la l\u00ednea celular acinar 266-6.
