Para mejorar el diagn\u00f3stico precoz, esfuerzos de investigaci\u00f3n se enfocan en caracterizar acontecimientos tempranos a la formaci\u00f3n del c\u00e1ncer, como lesiones preneopl\u00e1sicas y el origen celular del tumor. Las prote\u00ednas polycomb constituyen una familia de silenciadores epigen\u00e9ticos que se encuentran en varios tumores s\u00f3lidos. La hip\u00f3tesis principal es que polycomb desempe\u00f1a un papel en estados preneopl\u00e1sicos en el p\u00e1ncreas y en el desarrollo y progresi\u00f3n tumoral. La expresi\u00f3n de bmi1 y de ringb fue analizada durante el desarrollo pancre\u00e1tico, en tejido pancre\u00e1tico de modelos murinos y en muestras humanas de tejido pancre\u00e1tico. Se estudi\u00f3 el mecanismo de bmi1 utilizando modelos in vitro y con depleci\u00f3n inducida de bmi1. Bmi1 y ring1b se expresaron en precursores pancre\u00e1ticos exocrinos durante el desarrollo temprano y en c\u00e9lulas ductales y del islote, pero no en c\u00e9lulas acinares, en el p\u00e1ncreas adulto. Bmi1 se indujo en c\u00e9lulas acinares durante lesi\u00f3n aguda, en lesiones metapl\u00e1sicas acinoductales, en neoplasias intraepiteliales pancre\u00e1ticas (panin) y en pdac. En cambio, ring1b se indujo significativamente en panins de grado tard\u00edo y en pdac. La depleci\u00f3n de bmi1 en l\u00edneas celulares acinares cambi\u00f3 la expresi\u00f3n de los enzimas digestivos pancre\u00e1ticos.
