La replicaci\u00f3n tiene lugar tanto durante la fase s mit\u00f3tica como en la fase s premei\u00f3tica y requiere el ensamblaje del complejo de reconocimiento del origen (orc) en m\u00faltiples sitios del genoma denominados or\u00edgenes de replicaci\u00f3n (kong &#038; depamphilis, 2001). durante la meiosis la recombinaci\u00f3n se inicia a partir de roturas de doble cadena (dsbs) generadas tras la replicaci\u00f3n (cervantes et al, 2000). La reparaci\u00f3n de estas roturas genera crossovers que permiten la correcta segregaci\u00f3n de los cromosomas hom\u00f3logos y la generaci\u00f3n de diversidad en la progenie. Los dsbs son generados en todos los eucariotas estudiados por la prote\u00edna spo11 (rec12 en s. Pombe) por un mecanismo de transesterificaci\u00f3n tipo topoisomerasa (keeney et al, 1997). Tanto los eventos de recombinaci\u00f3n como la formaci\u00f3n de las roturas se producen en sitios espec\u00edficos del genoma llamados hotspots de recombinaci\u00f3n. Se ha comprobado que la mayor\u00eda de los hotspots en s. Pombe se sit\u00faan en regiones interg\u00e9nicas excepcionalmente largas de los cromosomas (cromie et al, 2007). mediante electroforesis bidimensional y southern blot hemos demostrado que existe un 60% de colocalizaci\u00f3n entre los or\u00edgenes de replicaci\u00f3n y los sitios de dsbs. Sin embargo tras varias inserciones y deleciones en dos loci diferentes del genoma, comprobamos que dicha colocalizaci\u00f3n no implica una dependencia funcional entre ambos procesos. tambi\u00e9n estudiamos cu\u00e1les eran los requerimientos necesarios para la generaci\u00f3n de dsbs integrando diferentes secuencias en dos loci gen\u00f3micos y observamos que el alargamiento de una regi\u00f3n interg\u00e9nica corta carente de dsbs genera roturas independientes de la actividad de un origen de replicaci\u00f3n adyacente o de la secuencia de la regi\u00f3n
