Vectores m\u00e1s eficientes, m\u00e1s sofisticados y con posibilidades nuevas, son el frente de avance y motor del campo. La presente tesis doctoral describe el desarrollo de dos nuevos vectores para terapia g\u00e9nica no viral, pertenecientes a \u00e1mbitos de aplicaci\u00f3n muy diferenciados. El lpp (lipopoliplejo) es un vector de asociaci\u00f3n liposomas\/adn\/pei cuya caracter\u00edstica esencial es la capacidad para transportar en compartimentos independientes una gran carga de ant\u00edgenos y genes u otras mol\u00e9culas, que son transferidos conjuntamente a la misma c\u00e9lula, y cuyo \u00e1mbito de aplicaci\u00f3n m\u00e1s apropiado ser\u00eda la dispensaci\u00f3n de ant\u00edgenos en estrategias terap\u00e9uticas de inmunizaci\u00f3n. Adicionalmente, he desarrollado varios m\u00e9todos nuevos  que permiten una caracterizaci\u00f3n completa de los vectores de asociaci\u00f3n liposomas\/policati\u00f3n\/adn, desarroll\u00e1ndose un protocolo que permite una caracterizaci\u00f3n completa de estos vectores previa a su uso en ensayos de inmunizaci\u00f3n.  Por otro lado, se han dise\u00f1ado vectores basados en tensoactivos dimerizables, dotados de una cabeza polar de dos amino\u00e1cidos que proporcionan las funciones qu\u00edmicas tiol y amino, y una cola hidrof\u00f3bica de diez carbonos de longitud; que son capaces de compactar monomolecularmente un pl\u00e1smido de adn, consiguiendo part\u00edculas de tama\u00f1o nanom\u00e9trico que constituyen el primer paso fundamental en la construcci\u00f3n de un vector de terapia g\u00e9nica no viral con estructura y tama\u00f1o semejantes a las de los virus. Los vectores desarrollados se han sometido a diversos ensayos biol\u00f3gicos, en particular en modelos de inmunizaci\u00f3n antitumoral en rat\u00f3n contra un ant\u00edgeno lip\u00eddico de melanoma (el gangli\u00f3sido de membrana gd3), y tambi\u00e9n con ant\u00edgenos proteicos hidrosolubles de la membrana de c\u00e9lulas tumorales de melanoma, obteni\u00e9ndose en ambos casos respuesta
