Los efectos de la adiponectina sobre sus tejidos diana depende muy posiblemente de la distribuci\u00f3n de sus formas moleculares. pioglitazona y rosiglitazona, agonistas del receptor ppar-g y cl\u00e1sicamente usados en el tratamiento de la diabetes tipo 2, tienen efectos sobre la v\u00eda de la adiponectina, pero poco se conoce sobre los efectos sobre la distribuci\u00f3n de sus formas moleculares y la expresi\u00f3n de sus receptores. el objetivo de esta tesis profundizar en el conocimiento de las formas moleculares de adiponectina y sus receptores y del efecto que los agonistas ppar-g pudieran tener sobre ello. Para la realizaci\u00f3n de este objetivo, se ha planteado como modelo de estudio a la rata zucker obesa, modelo de obesidad que presenta hiperinsulinemia y resistencia a la insulina. los resultados de este estudio indican que ambos medicamentos: a) tienen efectos insulinosensibilizadores parecidos, disminuyen el \u00edndice homa y los niveles plasm\u00e1ticos de insulina y \u00e1cidos grasos; b) aumentan el transporte de glucosa estimulado por insulina y el contenido total de glut- 4 en el m\u00fasculo esquel\u00e9tico; c) producen un cambio en la distribuci\u00f3n de las formas moleculares de adiponectina en plasma que se asocia a cambios en \u00edndice de sensibilidad a adiponectina (sa) pero no a cambios en los niveles de las formas moleculares; d) aumentan la expresi\u00f3n del receptor adipor1 en el m\u00fasculo esquel\u00e9tico y que rosiglitazona aumenta tambi\u00e9n la expresi\u00f3n del receptor adipor2 en el h\u00edgado.  obesity is nowadays a major medical problem that produces many diseases that cluster round insulin resistance, like the metabolic syndrome, type 2 diabetes mellitus and cardiovascular diseases. adiponectin, a novel protein characterized for having antidiabetic, antiinflammatory and anti-atherogenetic properties could be a new approach to understand and to treat these diseases. adiponectin acts through both its different molecular forms and its receptors. The adiponectin receptors are present in insulin sensitive tissues, like muscle, liver and adipose tissue. The effects of adiponectin on its target tissues could depend on the adiponectin molecular forms distribution. pioglitazone and rosiglitazone, ppar-g receptor agonists, are compounds classically used in the treatment of type 2 diabetes mellitus. These compounds act on adiponectin route, but little is known about their effect on the adiponectina molecular forms distribution and its receptors. the aim of this thesis is to have a more insight on the effect of the pparg agonist on adiponectina molecular forms distribution and on adiponectina receptors. To address this goal we have study the zucker fatty rat, a model of obesity with hyperinsulinemia and insulinresistance. the results of this study indicate that both drugs: a) produce similar insulin sensitizing effect, the homa index, plasma insulin levels and plasma free fatty acids levels are decreased; b) increase insulin-stimulated glucose transport and total glut-4 content in skeletal muscle; c) modify adiponectin molecular forms distribution in plasma that is associated with changes in the aiponectin sensitivity index (sa) but not with changes in the plasma levels of the molecular forms; d) increase adipor1 receptor expression in skeletal muscle and rosiglitazone also increases adipor2 receptor expression in liver.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
