Las c\u00e9lulas del estroma provienen de la c\u00e9lula stem mesenquimal cuyo papel en la fisiopatolog\u00eda de los s\u00edndromes mielodispl\u00e1sicos es controvertido. El objetivo de este trabajo es estudiar las c\u00e9lulas mesenquimales de estos pacientes. comenzamos con su caracterizaci\u00f3n. Se realiza la expansi\u00f3n de las c\u00e9lulas y estudios de citometr\u00eda y diferenciaci\u00f3n. Las mesenquimales de los pacientes tardan m\u00e1s tiempo en alcanzar confluencia, adem\u00e1s el n\u00famero de pases que soportan es menor. En el fenotipo presentan infraexpresi\u00f3n de cd105 y cd104. Se diferencian a adipocito, osteoblasto y condrocito, pero su patr\u00f3n de diferenciaci\u00f3n a esta \u00faltima est\u00e1 alterado. a continuaci\u00f3n se estudiaron las caracter\u00edsticas gen\u00e9ticas de las c\u00e9lulas, primero por hibridaci\u00f3n in situ y segundo por arrays de hibridaci\u00f3n gen\u00f3mica comparada. Por hibridaci\u00f3n encontramos que el porcentaje de c\u00e9lulas con la alteraci\u00f3n es del 10-26%, por lo tanto la mayor\u00eda de las c\u00e9lulas no ser\u00edan clonales. Sin embargo todas las c\u00e9lulas presentan alteraciones gen\u00f3micas. Adem\u00e1s las mesenquimales del s\u00edndrome 5q- presentan un perfil gen\u00f3mico diferenciado. \tfinalmente se valoraron las implicaciones en la fisiopatolog\u00eda de la enfermedad. Se valor\u00f3 la expresi\u00f3n mediante pcr de genes encontrados en el estudio de arrays gen\u00f3micos. Tambi\u00e9n se realiz\u00f3 estudio de microarrays de expresi\u00f3n. Y por co-cultivo se valor\u00f3 la potencial influencia de las c\u00e9lulas hematopoy\u00e9ticas sobre las mesenquimales. Por pcr se detect\u00f3 sobre expresi\u00f3n de los genes fgf4 e il32 en las mesenquimales del s\u00edndrome 5q-. Por microarrays de expresi\u00f3n se detect\u00f3 que las mesenquimales de este s\u00edndrome presentan un perfil de expresi\u00f3n diferente. En el co-cultivo se encontr\u00f3 que las hematopoy\u00e9ticas no influencian a las mesenquimales.  por lo tanto las alteraciones son intr\u00ednsecas a las c\u00e9lulas stem mesenquimales estando
