The role of the cell adhesion molecule psa ncam in cortical development

Psa is expressed in young neurons of the prenatal cerebral cortex of rodents (seki and arai, 1991) that are regarded today as tangentially migrating interneurons. Previous results have demonstrated the expression of psa-ncam in migrating interneurons of the rostral migratory stream and in fiber systems such as the mossy fibers of the hippocampus (rousselot et al., 1995; rutishauser and landmesser, 1996; cremer et al., 1997; seki and rutishauser, 1998). Important developmental alterations in these two systems have been described in vertebrates due to the absence of psa-ncam. According to these data and taking into consideration some pilot observations from our group, we determined to study the role of ncam on the development of the cerebral cortex using mice deficient for all nca isoforms (cremer et al., 1994). in this thesis i describe the spatio-temporal patterns of expression of psa and ncam in the telencephalon during the period of embryonic development. I also report alterations found in ncam null animals that are consequences of the lack of this protein. Such observations point to ncam as a molecule that plays certain roles in tangential migration of interneurons and in the proper establishment of thalamocortical projections.
